The overarching vision is to acquire the complete understanding of QC and modulate towards protein/cell health. This requires a choice: to focus on two dissimilar QC-‘client’ proteins, CFTR (LoF) and a-Synuclein (GoF), and provide a comprehensive overview of their fates in cells and the factors and conditions that shape it. A mechanistic understanding of key triage decisions requires their biochemical reconstitution and molecular analysis. This fundamental and complete understanding of the QC network forms the basis for future pharmacological intervention in protein diseases.
